Responsibilities This senior level technician will work independently under the direction of the principal investigator and scientific staff to design experiments; conduct literature reviews to assist in determining most suitable methods to be used in research and examine proposals to determine projects and methodologies best suited to research objectives and the overall goals of the laboratory. The incumbent will summarize and write for publication or presentation the results of research projects. The research technician will be involved in all aspects of murine preclinical studies in the Hingorani Laboratory, including but not limited to breeding, genotyping, ultrasound and other imaging techniques, and performing necropsies to obtain tissues for histology and cell lines. The technician may be involved in derivation, maintenance and study of primary cell lines; isolation of DNA, RNA, and proteins; as well as other techniques such as FACS sorting and expanding cells in vitro, analyzing cell phenotype and function using techniques including flow cytometry, proliferation assays, cytotoxicity assays, PCR, RT-PCR, sequencing, and ELISpot. Molecular techniques used in the lab include cloning, transfection/transduction of cell lines, gene targeting and northern and Southern blotting. Biochemical and cellular techniques include SDS-PAGE, western blotting, immunofluorescence, and immunohistochemistry. The technician will also assist with the management and organization of a fast-paced lab, and perform other duties as assigned by supervisor. Qualifications A Bachelor's degree in a biological field and at least 5 years of laboratory research experience including murine husbandry, cell tissue culture and molecular biology techniques are required Master's degree in a biological field and at least 2 years of experience in the techniques listed above are highly desired Computer skills: Microsoft Excel, Prism (Graphpad), FlowJo, and statistical programs for data analysis Excellent organizational and interpersonal skills are required Strong troubleshooting skills and the ability to multi-task with precise attention to detail is especially critical Experience with genetically engineered murine models of pancreas cancer or other solid tumors is desired Our Commitment to Diversity We are committed to cultivating a workplace in which diverse perspectives and experiences are welcomed and respected. Responsibilities The McGuire Lab (https://research.fhcrc.org/mcguire) is seeking outstanding postdoctoral fellows to work on a project involving the validating and testing of novel vaccine candidates to elicit broadly neutralizing antibodies against HIV-1. The successful candidate will design and execute prime-boost immunization regimens in a humanized murine model which will serve as a surrogate for human vaccine trials. The candidate will perform endpoint analyses including antigen-specific B cell sorting and serum antibody binding and neutralization assays to gain a high-resolution understanding of the immune response to immunization. To complement this analysis the candidate will perform single B cell sorting of human PBMC samples ascertain the identity of antigen-reactive nave B cells. Qualifications PhD in immunology, microbiology, cell biology, or a related field is required and expertise in B cell immunology is highly desirable. Experience with conducting in vivo studies in murine models, preferably immune analysis, required. The candidate should have a strong molecular biology background as well. Must be willing to work flexible hours as needed. The candidate must be independent, show initiative, willingness to take up new skills and responsibilities, and be a team player. Excellent attention to detail and the ability to work independently are essential to success in this position, as are good communication and organizational skills. The candidate must be self-motivated, able to multitask and to work within a very collaborative environment.
